Into the OD channel, amp gain on 3-4, Pedal drive on 0, level on full.
But lately I've just been using the ease of the GT 10's Tscreamer into my OD channel and amp control to switch to clean.
I'll turn the amp gain down now and then depending on the music.

One overdrive, one distortion. Maybe a decent fuzz (Fuzz Face or Big Muff) for those so inclined (I'm not). Any good models - a Tubescreamer and a Rat (or boutique versions therof) covers most bases in my experience. Everything else is fairly subtle variation, and I find it's usually best to work at getting the best out of what you've got (including the amp) than chopping and changing, searching for the perfect tone out the box.
My Amps are the Vox AC15 Hand Wired and a '65 Fender Twin (original) - very different animals.
I've been using a Vox Big Ben with a high quality NOS valve (Philips goldpin IIRC) the last year or so which has a nice, neutral overdrive tone and can kick the amp into higher gain nicely, but doesn't do higher gain stuff and is a bit of a one trick pony (although the same could be said for most pedals and it does do that trick very well).
Currently testing the TC Electronic Nova Drive and think I'll invest in one of those as my go-to. It's basically a TS (with a less of a midrange hump) and a Rat (with a bass and treble instead of the filter control) in one box. The nice thing is the control - you can stack them or switch between them, swap the order of the two or even put them in parallel. And save all the settings to patches. Surprisingly good sound quality, easy to use and true bypass.

here is mine:
Boss SD-1 through a Peavey Delta Blues(30 watt into 16ohm 15'' speaker).
I play various styles from soft blues to zakk wylde pinching metal.
I had the pleasure of testing out the tube tamer this week with my amp cranked to nearly max, and did some experimenting with pedals.
SD-1 through clean channel:
Gives a great Eddie Van Halen type rhytmn sound exactly like on the recording of their song"cant stop loving you", without the acoustic guitar and hyped up chorus effect.
Can be used to give a very dynamic sound when playing to add lots of soul to the track.
SD-1 through pre amp channel:
This is where this little beast comes alive, depending on the pre and post settings one can easily go from def lepard to some of 90's albums from ozzy featureing zakk wylde.
The EQ settings on the amp can also contribute alot to tuneing in that sound you want.
I have a boss DS-1 that i'll give a go tonight, because i haven't used it in a while, maybe my perspetion of sound may have changed over the past few months.
PeteM@ What style are you playing?

Hmmm well depending on music style but overall i definately LOVE my MAXON OD808
BLUES/ROCK (Think SRV or Hendrix'y altho i like a little more gain/sustain)
Fender MIA STRAT / EPI LES PAUL > maxon od808 >peavey classic 30 > mesa rectifier 2x12 cab
(here i like to run the gain full and sometimes another tubescreamer infront of it at about 1/4 gain just for alittle more)
METAL/HARD ROCK (think Killswitch engage or Bullet for my valentine or As i lay dying - get all these tones pretty much)
Ibanez loaded with seymour duncan JB / cort loaded with EMG 81 > maxon od808 > peavey 6505+ > mesa rectifier 2x12 cab
(often swop from running the gain on 0 or about 1/4 way up)
JoeyBones
I love my Radial Bigshot PB1 and the booster on Box Of Rock. They stack very nicely together.

Digitech Hardwire range seems VERY COOL and true bypass ?
looks like solid pedals with ALOT of thought behind them and when i tested one through an orange it sounded MASSIVE
PeteM
@Manfred - blues and rock - not metal

@Manfred - blues and rock - not metal
then you need something low wattage and maybe just one overdrive pedal.
If you have narrowed it down to a few amps and pedals you might want to own oneday, go check out youtube there's loads of videos with gear demo's.
Also try and keep in budget, some amps can get expensive.
Also test amps out at your local musc shop, to train your ear how to listen to what sounds best for you.

@ Mannie
What about the clean tones of Mark Knopfler and Robert Cray. I can almost get these sounds with my Peavey T60.
But I am using my POD through the PA System. Will a tube amp improve the sound? Or do I need to acquire a Fender Strat as well?

You using a compressor? Fairly important to many of the early Dire Straits Knopfler stuff.
The T-60's are great guitars - very innovative and capable of getting quite a wide range of tones from fatter, chunkier stuff through skinny single-coil sounds (when the tones are turned up) and funky OOP (Out Of Phase) sounds, but they are all fairly unique tones. It'll do a reasonable facsimile of many sounds, but never quite nail Strat or Les Paul tones IMO.
Manfred-Klose
you dont need to spend money on a strat if you dont want to, squiers strats is alot cheaper and also gets the job done.
a pod through a PA system works fine, chances are you can get more popular tones this way, aldough adding a tube amp can give you some dynamics and warm up the signal.
So its all down to experimenting and see what works for you.

Does that Maxon really make the tone better for the 5150? Ive got one but never used it live.. never liked the tone much? But then Ive never tried it with low amp gain and booster pedal..

I've tested out the old boss DS-1 last night again.
Through the clean it gives a very bright and "happy(70's type)" drive sound(bad company ??)
Through the pre amp channel it sounds like the earlier stuff from joe satriani.
It's to trebly for me.

On the 5150 do you you use the rhythm channel or lead channel?
I find the lead channel a bit 'thinner' ?
MIKA-the-better-one
sIMPLE a ts808 or clone OR BD2/ mi audio blues pro into a proco rat or a fuzz face or a good clone (mxr fuzz 108) into a fender amp (tube)
