How hard is it to get a setup where a note played on a string sounds exactly the same as the exact same note played on an different string? (same octave, obviously). Is this desirable? Impossible?

You mean say an F# on the 1st (2nd fret) sounding exactly the same as the F# on the 2nd (7th fret)?

I don't think that's possible, because of the different tensions.

Generally notes physically higher up the neck have a different characteristic from those lower down.

Yar, thats exactly what I mean. Ive been doing some experimentation with octave jumps but preserving certain parts of the riff, octave-wise, but it never sounds the same on a different string. Generally the thicker strings have a much smoother tonal quality.

it also depends on the importance of the note being played.. is the note emphasised or just a passing note..

I just re-tabbed something because my little stubby sausage fingers do not have the reach to comfortable get to certain notes when I have to, so I have re-tabbed to playing them on a different string, closer to where the current action is.. luckily they are just passing notes, so the tonal differences are not that noticeable.
